Monoclonal Antibody Generation Services Using the ADLib® System

The ADLib® system, a proprietary monoclonal antibody generation technology developted by Chiome, supports the generation of antibodies essential for antibody therapeutics, in vitro diagnostics and life science research.

ADLib® Service Lineup

ADLib®mAb generation ADLib®Affinity Maturation
(KI-AMP)
Type Monoclonal antibody generation Affinity maturation
Overview Antigen-specific antibody generation service using ADLib® libraries Affinity improvement services for existing antibodies (e.g. hybridoma-derived antibodies).
Period About 3 weeks to report ELISA screening results. About an additional 2 weeks until reporting of antibody sequence information for candidate clones. About 8 weeks to construct a strain expressing desired-existing antibodies, involving all steps from vector construction to cell line QC, where the target antibody sequence is introduced into the ADLib® system. This is followed by Affinity maturation for around 1.5 months (including diversification, screening, sequencing and SPR analysis).
Antigen types

  • Peptides
  • Antibodies (anti-idiotypic antibodies)
  • Low molecular weight compounds

Advantages
  • Rapid results.
  • Completely in vitro system, no use of animals.
  • Selection and screening using full body IgG.
  • Efficient selection using a cell sorter.
  • Human antibodies can be obtained directly (no need for humanisation)
  • The method is based on inducing mutations in the antibody genes within DT40 cells (proprietary technology) and the isolation of candidate clones that show a higher affinity to the antigen.
  • The original technology has been further improved to achieve a shorter timeframe and lower cost compared to our conventional method.

Comparison with Other Antibody Generation Platforms

The ADLib® system is a method for selecting antigen-specific antibodies from a library of full-length antibodies displayed on the cell surface. Although phage display is widely recognized as the same platform, it differs in technical characteristics across various aspects (refer to the table below). In addition, the construction of libraries and the screening process often rely on the unique know-how and technology developed by individual laboratories.

Other well-established methods, such as hybridoma and B-cell cloning, exhibit numerous differences from the cell-surface display. For example, the immunisation process is necessary.
